Apoptosis-associated speck-like protein containing a caspase activation and recruitment domain (ASC), an adaptor protein for inflammasome receptors, is essential for inducing the caspase-1 activaton and the consequent secretion of IL-1?, which associates with local inflammation during liver ischemia/reperfusion injury (IRI). However, little is known on mechanisms by which ASC/Caspase-1/IL-1? axis exerts its function in hepatic IRI. This study was designed to explore the functional roles and molecular mechanisms of ASC/Caspase-1/IL-1? signaling in the regulation of inflammatory responses in vitro and in vivo. Using a partial lobar liver warm ischemia (90min) model, ASC-deficient and WT mice (C57BL/6) were sacrificed at 6h of reperfusion. Separate animal cohorts were treated with anti-IL-1? Ab or control IgG (10mg/kg, day -1, i.p.). We found that ASC deficiency inhibited Caspase-1/IL-1? signaling, leading to the protection against liver IR-damage, local enhancement of antiapoptotic functions, and downregulation of HMGB1-mediated TLR4-driven inflammation. Interestingly, treatment of ASC-deficient mice with rHMGB1 recreated liver IRI. Moreover, neutralization of IL-1? ameliorated the hepatocellular damage by inhibiting NF-?B/COX2 signaling in lR-stressed livers. In parallel in vitro studies, knockout of ASC in LPS-stimulated bone marrow derived-macrophages depressed HMGB1 activity via p38 MAPK pathway, leading to the inhibition of TLR4/NF-?B, and ultimate depression of proinflammatory cytokine programs. Conclusion: ASC-mediated Caspase-1/IL-1? signaling promotes HMGB1 to produce TLR4-dependent inflammatory phenotype, leading to hepatocellular injury. Hence, the ASC/Caspase-1/IL-1? signaling mediates inflammatory response by triggering HMGB1 induction in hepatic IRI. Our findings provide the rationale for a novel therapeutic strategy to manage liver injury due to IR. (HEPATOLOGY 2013.).

Factors related to marital satisfaction in women with major depressive disorder.

Major depressive disorder (MDD) is one of the most common psychiatric disorders which affects married couples frequently.The present study aims to explain the role of family processes, social support and demographic factors in marital satisfaction of women with Major Depressive Disorder (MDD).In this cross-sectional study, 188 women with MDD were randomly selected among the patients who visited Bozorgmehr Clinic of Tabriz University of Medical Sciences. The sample selection was carried out through structured psychiatric interviews based on DSM-TV-TR criteria. Data were collected using Index of Marital Satisfaction (IMS), Family Process Scale (FPS) and Norbeck Social Support Questionnaire (NSSQ).The Mann Whitney U, Multivariate and ANOVA tests were used to analyze the data.No relationship was observed between age, educational level, age difference of couples and number of children with family processes and marital satisfaction (p ? 0.05). The patients with low educational level reported less social support (p ? 0.05).Marital satisfaction and family coherence were lower when the husband had a psychiatric disorder (P ? 0.01). The family processes (family coherence, problem-solving skills, communication skills and religious beliefs) and social support positively predicted marital satisfaction, while the husband’s psychiatric disorders negatively predicted marital satisfaction.The findings highlight the significance of family processes, social support and husband’s psychiatric disorders in marital satisfaction of women with MDD.

Individual, family, social, and cultural predictors of depressed mood in former soviet immigrant couples.

Gender differences in predictors of depression for married couples from the former Soviet Union were examined in a cross-sectional, descriptive analysis. Data were collected during a longitudinal study of post-migration health and adaptation. The sample included 308 men and women (154 couples), ages 40-79, who had lived in the US for an average of 6 years. Generativity, marital satisfaction and communication, social support, immigration challenges, and alienation were independent predictors of depressed mood. A gender interaction was found for generativity, indicating that diminished opportunities to guide the next generation and be productive members of society may have been more depressing for women. Interventions should attend to gender differences in developmental needs, reduce immigration-related challenges, and strengthen family and social support. Published 2013.
