Comparative phylogeography can elucidate the influence of historical events on current patterns of biodiversity and can identify patterns of co-vicariance among unrelated taxa that span the same geographic areas. Here we analyze temporal and spatial divergence patterns of cloud forest plant and animal species and relate them to the evolutionary history of naturally fragmented cloud forests-among the most threatened vegetation types in northern Mesoamerica. We used comparative phylogeographic analyses to identify patterns of co-vicariance in taxa that share geographic ranges across cloud forest habitats and to elucidate the influence of historical events on current patterns of biodiversity. We document temporal and spatial genetic divergence of 15 species (including seed plants, birds and rodents), and relate them to the evolutionary history of the naturally fragmented cloud forests. We used fossil-calibrated genealogies, coalescent-based divergence time inference, and estimates of gene flow to assess the permeability of putative barriers to gene flow. We also used the hierarchical Approximate Bayesian Computation (HABC) method implemented in the program msBayes to test simultaneous versus non-simultaneous divergence of the cloud forest lineages. Our results show shared phylogeographic breaks that correspond to the Isthmus of Tehuantepec, Los Tuxtlas, and the Chiapas Central Depression, with the Isthmus representing the most frequently shared break among taxa. However, dating analyses suggest that the phylogeographic breaks corresponding to the Isthmus occurred at different times in different taxa. Current divergence patterns are therefore consistent with the hypothesis of broad vicariance across the Isthmus of Tehuantepec derived from different mechanisms operating at different times. This study, coupled with existing data on divergence cloud forest species, indicates that the evolutionary history of contemporary cloud forest lineages is complex and often lineage-specific, and thus difficult to capture in a simple conservation strategy.

Proteomic analysis of lymphoblastoid cells derived from monozygotic twins discordant for bipolar disorder: a preliminary study.

Bipolar disorder is a severe mental illness characterized by recurrent manic and depressive episodes. In bipolar disorder, family and twin studies suggest contributions from genetic and environmental factors; however, the detailed molecular pathogenesis is yet unknown. Thus, identification of biomarkers may contribute to the clinical diagnosis of bipolar disorder. Monozygotic twins discordant for bipolar disorder are relatively rare but have been reported. Here we performed a comparative proteomic analysis of whole cell lysate derived from lymphoblastoid cells of monozygotic twins discordant for bipolar disorder by using two-dimensional differential in-gel electrophoresis (2D-DIGE). We found approximately 200 protein spots to be significantly differentially expressed between the patient and the co-twin (t test, p<0.05). Some of the proteins were subsequently identified by liquid chromatography tandem mass spectrometry and included proteins involved in cell death and glycolysis. To examine whether these proteins could serve as biomarkers of bipolar disorder, we performed Western blot analysis using case-control samples. Expression of phosphoglycerate mutase 1 (PGAM1), which is involved in glycolysis, was significantly up-regulated in patients with bipolar disorder (t test, p<0.05). Although PGAM1 cannot be regarded as a qualified biomarker of bipolar disorder from this preliminary finding, it could be one of the candidates for further study to identify biomarkers of bipolar disorder. Psychometric properties of the arabic version of the obsessive compulsive beliefs questionnaire-44 in a student population.

We examined the psychometric properties of the Arabic version of the Obsessive Compulsive Beliefs Questionnaire-44 (OBQ-44) in a sample of Kuwait University students. This questionnaire was developed by the Obsessive Compulsive Cognitions Working Group in order to assess belief domains believed to be crucial in the development of obsessive compulsive symptoms.The Arabic version of the OBQ-44 was developed according to the standard translation and back-translation methods. The Arabic versions of the OBQ-44, the Maudsley Obsessive – Compulsive Inventory (MOCI), and Beck Depression Inventory-Revised (BDI-II) were then administered on a sample of 200 Kuwait University students from the faculty of humanities chosen through random cluster sampling. Retest was administered within a 4 week time period.The results of principle component factor analysis with varimax rotation indicated 6 factors which overlapped to a high degree. A 3 factor solution was chosen based on the scree plot and factor loadings which explained 36.12% of the variance. The factors were labeled as responsibility and threat estimation (RT), importance and control of thought (ICT) and perfectionism/Certainty (PC). The reliability coefficient of the three factors and the total score were assessed using three methods: Internal consistency, Test-retest reliability and Split-half reliability. Results showed an acceptable internal consistency for the Arabic version of the OBQ-44. Regarding the validity of OBQ-44, the instrument correlated with the total score of MOCI and most of its subscales.These data support the reliability and validity of the OBQ-44 in a sample of Kuwait University students.
