Persistent maladaptive memories that maintain drug seeking and are resistant to extinction are a hallmark of addiction. As such, disruption of memory reconsolidation after retrieval has received attention for its therapeutic potential. Unrestrained reconsolidation may have the opposite effect, leading to reiterative and cumulative strengthening of memory over long periods of time. Here we review the molecular mechanisms underlying reconsolidation of appetitive and drug-rewarded memories, and discuss how these findings contribute to our understanding of the nature of this process. Finally, we suggest that drug-induced alterations to signal transduction might lead to dysregulation of reconsolidation, causing enhancements of drug-related memory after retrieval, and significantly contribute to the compulsive drug seeking that is a core component of addiction.

The relationship between optimal parenting, Internet addiction and motives for social networking in adolescence.

This paper presents a cross-sectional study of a large, high-school Greek student sample (N=1971) with the aim to examine adolescent motives for participating in social networking (SN) for a possible link with parenting style and cognitions related to Internet addiction disorder (IAD). Exploratory statistics demonstrate a shift from the prominence of online gaming to social networking for this age group. A regression model provides with the best linear combination of independent variables useful in predicting participation in SN. Results also include a validated model of negative correlation between optimal parenting on the one hand and motives for SN participation and IAD on the other. Examining cognitions linked to SN may assist in a better understanding of underlying adolescent wishes and problems. Future research may focus in the patterns unveiled among those adolescents turning to SN for the gratification of basic unmet psychological needs. The debate on the exact nature of IAD would benefit from the inclusion of SN as a possible online activity where addictive phenomena may occur.

Connection of toxin-antitoxin modules to inoculation eschar and arthropod vertical transmission in Rickettsiales.

The biological role of toxin-antitoxin systems (TAS) in pathogenicity and cell addiction of Rickettsia was recently reported. We realized a comparative genomic analysis onto 33 rickettsial genomes and correlated the presence of TAS encoding genes with vertical transmission (VT) in arthropod hosts, the presence of inoculation eschar in humans and experimental animals, and the mortality in humans. There is a significant statistical link between TAS and the presence of an eschar (p?0.0001). The presence of TAS is also significantly inversely correlated with mortality. The toxic effect of TAS may increase the local reaction, thus inhibiting the spread of rickettsiae associated with fatal outcome of the disease. The TAS were also linked to VT (p?0.0001). Together with our previous findings we speculate that this is the first addiction system evidenced in intracellular bacteria. Thus, the TAS, as selfish genetic elements, might be essential to the evolutionary strategy of intracellular bacteria.
